Tamoxifen metabolism pathway in humans [16]. CYP 2D6 is the major isoform responsible for the formation of 4-hydroxytamoxifen and CYP3A4/5 is the major isoform responsible for the formation of N-desmethyltamoxifen, whereas the generation of Endoxifen is predominantly catalyzed by CYP2D6. Other CYP isoforms, including CYP2C19, CYP2C9, and CYP2B6, appear to play less important roles in Tamoxifen metabolism in vitro at therapeutically relevant concentrations
